Abstract: | OBJECTIVE To discover the corelationship between nm23 expression and nucleoside diphosphokinase activity in gastric carcinoma.METHOD Homogenate,cytosol,microsome,mitochondria were obtained from gastric carcinoma and normal tissues and nucleoside diphosphokinase activity was determined by Kezdi's method. nm23 expression in those tissues was also studied by using nm23 H 1 monoclonal antibody with avidin biotin complex method. RESULT Nucleoside diphosphokinase activity(per gram tissue) and specific activity in both homogenate, in cancer group(265 99±84 81mU,2 70±0 95mU/mg protein) and cytosol(290 81±88 18mU,3 57±1 10mU/mg protein) are significently higher than those in normal group(181 28±63 61,229 57±64 67mU; 1 75±0 61,2 74±0 75mU/mg protein) Activities and specific activities in mitochondtia of both group has no statistical difference and in microsome fractions in normal group(8 89±5 84mU,1 92±0 88mU/mg protein) are a little higher than those in cancer group(5 70±2 60mU,1 37±0 61mU/mg protein). CONCLUSION The nucleoside diphosphokinase activity in nonmetastatic tissue is higher than that in normal tissue. |
