兔关节滑膜细胞的分离、培养和纯化

目的探讨兔关节滑膜细胞的体外分离、培养和纯化方法。方法无菌条件下,从6月龄新西兰白兔的膝关节囊内剪取滑膜组织,采用组织块培养法和酶消化法分离滑膜细胞。经台盼蓝拒染计数,将细胞按1×106接种于培养瓶,传代培养。结果经反复贴壁和多次消化达到形态学上的纯化要求,活性率为96%,纯度达95%以上,培养的细胞具有成纤维细胞形态和特征。结论本研究建立了简单易行的滑膜细胞分离、培养和纯化方法。

Isolation, cultivation and purification of rabbit articular synovial cells

Under aseptic condition, the synovial membrane of knee joint from New Zealand white rabbits of 6 months of age was incised with a sharp knife, and the synovial cells were isolated by means of tissue explant cultivation and enzyme digestion. The viable cells after counting with trypan blue dye exclusion were then transferred to culture flask containing DMEM medium in a density of 1×10 6. After repeated adherence and enzyme digestion, the cells attained the requirement of morphological purification with a purity of more than 95%. These cultured cells exhibited 96% viability and showed the morphological characteristics of fibroblasts. This method of isolation, cultivation and purification of the synovial cells in vitro is proved to be simple and feasible.
