天麻钩藤颗粒对硝酸甘油致偏头痛模型大鼠的影响

摘    要:目的:观察天麻钩藤颗粒对硝酸甘油致偏头痛模型大鼠的影响。方法:将实验大鼠随机分为6组,正常对照组、模型对照组、西比灵胶囊组、天麻钩藤颗粒低、中、高剂量组。正常对照组和模型对照组灌服蒸馏水,阳性组灌胃给予西比灵胶囊(1×10-3g/kg),给药组灌胃给予相应浓度的天麻钩藤颗粒(低剂量:0.75g/kg,中剂量:1.5g/kg,高剂量:3g/kg)7d,于末次给药30min后,除正常对照组皮下注射生理盐水外,其余各组均皮下注射硝酸甘油注射液造模,并观察其耳红、挠头次数等行为学指标;取血,测定血液流变学,酶免法测定各组大鼠血清NO、SOD、MDA及脑组织肾上腺素(E)、去甲肾上腺素(NE)、5-羟色胺(5-HT)和多巴胺(DA)的含量。结果:天麻钩藤颗粒中、高剂量组能显著缩短模型大鼠耳红消失时间(P0.01),此作用呈一定的剂量依赖性。天麻钩藤颗粒高剂量组能显著减少模型大鼠挠头次数(P0.05,P0.01)。天麻钩藤颗粒高剂量组能显著降低模型动物全血黏度低、中、高切变率、血浆黏度切变率及全血还原黏度低、中切变率(P0.05,P0.01)。天麻钩藤颗粒中剂量组能显著降低模型动物血清MDA含量,升高SOD含量(P0.05),高剂量组能显著降低模型动物血清NO、MDA水平(P0.05),升高SOD含量。天麻钩藤颗粒低、中、高剂量组较模型大鼠脑组织E、NE、5-HT、DA含量均明显升高(P0.05,P0.01)。结论:天麻钩藤颗粒能预防偏头痛发作,这种作用可能与调节机体单胺类神经递质的释放和代谢机制得以恢复,进而改善脑及血管的功能障碍有关。

Influence of Tianma Gouteng Granules on migraine rat induced by nitroglycerin

Abstract:Objective: To observe Tianma Gouteng Granule’s(TMGTG) affection on rat model of migraine induced by nitroglycerin.Methods: All the rats were randomly divided into 6 groups,normal control group,model control group,flunarizine capsule group(1×10-3g/kg),Tianma Gouteng Granule low dose gourp(0.75g/kg),Tianma Gouteng Granule medium dose group(1.5g/kg),Tianma Gouteng Granule high dose group(3g/kg).The other groups were continuously given intragastrically Tianma Gouteng Granule for 7 days except the normal and model control group.The other groups were injected subcutaneously nitroglycerin induced migraine model except that the normal control group was injected subcutaneously normal saline 30min after the last administration.Red ears and scratching frequency of all the rats were considered as behavioral indicators,blood was taken to assay blood rheology and serum NO,SOD,MDA and brain epinephrine(E),norepinephrine(NE),5-hydroxytryptamine(5-HT) and dopamine(DA) through ELISA method.Results: The high dose group and medium group of TMGTG had significantly reduced the disappearance time of model rats red ear(P<0.01),this effect was a dose dependent manner.TMGTG high dose group can significantly reduce the number of scratching their heads of model rats P<0.05,P<0.01).TMGTG high dose group could significantly reduce the blood viscosity of low,medium and high shear rate,plasma viscosity shear rate and whole blood reduced viscosity of low,medium shear rate of model animal(P<0.05,P<0.01).TMGTG medium dose group significantly reduced serum MDA and increased SOD of model animal(P<0.05).High dose group of TMGTG could significantly reduce the serum NO,MDA levels(P<0.05) and increased SOD of model animal.Low,medium and high dose groups of TMGTG can significantly increase E,NE,5-HT,DA levels of brain tissue(P<0.05,P<0.01).Conclusion: Tianma Gouteng Granule can prevent migraine attacks,which may regulate the release of monoamine neurotransmitters and the metabolism of the body,thus improving the function of the brain and vascular disorders.
