识别血吸虫成虫多肽表位单克隆抗体细胞株的建立及用于检测循环抗原的研究

摘    要:作者获得1株特异的抗血吸虫成虫20-24kDa蛋白多肽表位的杂交瘤细胞株(SjA111)。分泌McAb为IgG1,腹水效价1.2×106。将SjA111McAb-HRP结合物用Dot-ELISA检测499例慢性血吸虫病患者的循环抗原(CAg),阳性者390例(78.16%);25例肺吸虫病人,36例华支睾吸虫病人及132例正常人血清中,仅1例肺吸虫病人出现交叉反应。CAg的检出率和阳性血清滴度倒数的几何均值(GMRT)随着每g粪便虫卵数(EPG)的增加而递增。吡喹酮治疗后各期病人粪孵阴性者CAg的转阴率:半年为75.50%(114/51),1年为73.68%(84/114),1年半为88.04%(103/117)。

Abstract:A hybridoma cell line (Sj A111) secreting McAb of 20-24kDa polypiptide epitopes from adult worm of S. japonicum has been prepared. The immunoglobulin class of SjA111 McAb was IgG1. The titer of ascited containing McAb reached 1.2×106. 499 cases with chronic schistosomiasis were detected by Dot-ELISA using SjA111 McAb-HRP conjugate. Positive rate of CAg was 78.15% (390/499). Only 1 out 25 cases with paragonimiasis showed the cross reaction while other 36 cases with clonorchiasis and 132 normal individuals showed negative reaction. The detection rate and GRMT of CAg corresponded with the increase of EPG. Six months, 1 year and 1. 5 years after effective treatment. The negative conversion rate of CAg became 75.5% (114/151), 73.69% (84/114) and 88. 0% (103/117) respectively.
