细胞间黏附分子-1在脂多糖致大鼠脑水肿中的表达

摘    要:目的 探讨脂多糖 (LPS)致大鼠脑水肿发生过程中细胞间黏附分子 1(ICAM 1)的表达与变化。方法 健康SD大鼠 10 5只 ,随机分为空白对照组 (A组 ,n =5 ) ,生理盐水组 (NS组 ,n =5 0 ) ,脂多糖组 (LPS组 ,n =5 0 )。LPS诱导大鼠脑水肿发生 ,制备脑组织冰冻切片 ,免疫组织化学染色方法测定ICAM 1的表达情况 ,图像分析仪测定ICAM 1阳性细胞单位面积的光密度值 (A值 )。结果 各时间点LPS组脑组织含水量、伊文思蓝 (EB)含量、ICAM 1表达情况及表达量均高于A组和NS组 ,差异有显著性 (P <0 .0 5 )。ICAM 1表达量和脑组织含水量、ICAM 1表达量和EB含量均呈正相关 (r =0 .5 37,0 .4 6 7,0 .5 4 9 P均 <0 .0 5 )。结论 LPS可诱导脑水肿发生 ,脑血管内皮细胞表面ICAM 1表达增强 ,促进血脑脊液屏障通透性的破坏和脑组织含水量增加 ,介导炎症反应过程

Abstract:Objective To study the expression of intercellular adhesion molecule 1(ICAM 1) in the brain edema induced by lipoposacchride(LPS) in rats.Methods Immunohistochemistry staining method was used to measure the expression of ICAM 1 in brain tissue after brain edema was induced by LPS in rats. Image pattern analyzer was used to determine the photometric density of ICAM 1 positive cell area.Results At each time point, the content of brain tissue,evans blue(EB) and the expressing quantity of ICAM 1 in LPS group increased significantly compared with group A and NS group(P<0.05). In LPS group, water content in brain tissues and EB content , expressing quantity of ICAM 1 and brain water content, expressing quantity of ICAM 1 and EB content all were positively correlated (r=0.537,0.467,0.549 P<0.05).Conclusions ICAM 1 could participate in the onset and development of brain edema induced by LPS, enhance the expression of ICAM 1 on cerebrovascular endothelial cell surface, damage the permeability of blood cerebral barrier, increase the water content of the brain tissues, and induce the reaction of inflammation.
