摘    要:目的:评价药物“911”在鸭实验动物模型中抗鸭乙型肝炎病毒(DHBV)的效果。方法:北京鸭足静脉注射DHBV,第1次试验随机分为空白,Ara-AMP对照和20,40,80mg/kg3个治疗组,于感染后第7d,给药后第5,10d和停药后第5d采血,第2次试验设空白对照和40.80mg/kg2个治疗组,用药后第5,10d及停药后第5d各组分别处死3只鸭取肝脏检测肝HDBV DNA,采用DOT EIA法检测血清DHBsAg,斑点分子杂交法检测DHBV DNA,Southern转膜杂交检测鸭肝DHBV DNA。结果:第1次试验结果表明,血清DHBsAg及DHBV DNA在20mg/kg组基本无变化,在40mg/kg组治疗后明显下降,但效果无80mg/kg组明显;80mg/kg组DHBsAg 及DHBV DNA水平治疗效果最明显,与用药前及空白对照比较均有统计学意义。第2次试验与第1次试验的结果基本吻合,鸭肝脏DHBV DNA检测表明,80mg/kg治疗组对鸭肝DHBV DNA复制有明显抑制作用。可减少鸭肝DHBV DNA含量。结论:“911”有较好的抗DHBV感染效果,具有明显的剂量反应关系和可重复性。

Abstract:Objective: To evaluate the anti DHBV effect of "911" in experimental ducks infected with DHBV. Methods: In one experiment, five groups of infected ducks were treated with saline, Ara AMP and "911" of 20,40,80mg/kg doses respectively. Blood was collected from the ducks on the 7?d after infection, on 5?d and 10?d during treatment and on 5?d after treatment. In another experiment, three groups of infected ducks were treated with saline and "911" of 40,80 mg/kg doses respectively. The ducks were killed and the liver and blood were collected on the 5?d and 10d during treatment and 5?d after treatment. Serum DHBsAg and DHBV DNA levels were detected by dot EIA and dot hybridization respectively, and liver DHBV DNA was measured by Southern blot hybridization. Results: The results of the 1st experiment showed that the serum DHBsAg and DHBV DNA did not change in the 20?mg/kg dosage group, but there was a significant drop in the 40?mg/kg and 80?mg/kg groups, especially in the 80?mg/kg group. The results of 2?nd experiment virtually coincided with those of the 1st one. Detection of DHBV DNA in the duck livers showed that the replication of DHBV DNA was inhibited and the amount of DHBV DNA declined in the 80?mg/kg group. Conclusion: "911" has excellent anti DHBV effect in ducks infected with DHBV and shows a distinct dose effect relationship and can be reliably repeated.
