莫诺苷对局灶性脑缺血再灌注大鼠Wnt7a和APC表达的影响

摘    要:目的观察莫诺苷对局灶性脑缺血再灌注大鼠Wnt7a和APC表达的影响。方法用线栓法制备大鼠局灶性脑缺血再灌注模型(MCAO)。将50只Sprague-Dawley大鼠随机分为假手术组,模型组,莫诺苷小、中、大剂量组(30 mg/kg、90mg/kg、270 mg/kg)。利用蛋白免疫印迹法检测Wnt7a和APC表达变化。结果造模7 d,模型组大鼠皮层Wnt7a的表达比假手术组显著升高,APC表达显著下降;与模型组相比,莫诺苷大剂量组Wnt7a表达明显升高;莫诺苷小、中、大剂量组APC的表达显著降低。结论莫诺苷可以促进缺血性脑损伤后Wnt7a表达,抑制APC表达,从而激活Wnt信号通路。

Abstract:Objective To investigate the effects of morroniside on the expression of Wnt7a and APC in rat cortex of focal cerebral ischemia/reperfusion. Methods After middle cerebral artery occlusion (MCAO) model was performed SD rats were divided into groups of sham, model, and morroniside (30 mg/kg, 90 mg/kg,270 mg/kg). The expression of Wnt7a and APC were evaluated by Western blot. Results Compared with the sham group, the expression of Wnt7a in rats of MCAO model was increased, and the expression of APC was decreased. Compared with model group, morroniside at the concentrations of 30 mg/kg, 90 mg/kg, 270 mg/kg decreased APC expression respectively, while 270 mg/kg morroniside-treatment obviously increased the expression of Wnt7a. Conclusion Morroniside could increase the expression of Wnt7a, and inhibit the expression of APC after ischemic injury, therefore activating the Wnt signaling pathway.
