摘    要:目的 采用多因素复合的造模方法,先建立湿热模型,采用改良Coderre1法痛风造模,以复制痛风湿热蕴结证病证结合动物模型2,探明水通道蛋白-2(AQP2)、热休克蛋白70(HSP70)在痛风湿热蕴结证中表达的意义,为模型建立指标评估及临床药物疗效判定指标做进一步实验验证。方法 实验对象为SPF级雄性SD大鼠,将40只大鼠按随机数字表分为4组,分别为正常对照组、痛风模型组、湿热模型组、痛风湿热模型组,每组10只。对各组分别造模(方法详见正文造模部分),造模期间观察大鼠外观、精神状态、食饮量、体重变化等一般情况,测定受试踝关节炎症指数、造模72 h各组大鼠的步态情况、血尿酸、尿液AQP2、血浆HSP70等指标。结果 4组模型尿液AQP2含量具有显著差异;与正常对照组比较,湿热模型组、痛风湿热模型组尿液AQP2明显升高(P < 0.05;P < 0.05);与痛风模型组对比,湿热模型组、痛风湿热模型组尿液AQP2明显升高(P < 0.05;P < 0.05)。四组模型尿液HSP70含量具有显著差异;与正常对照组比较,湿热模型组、痛风湿热模型组血浆HSP70明显升高(P < 0.05;P < 0.05);与痛风模型组对比,湿热模型组、痛风湿热模型组血浆HSP70明显升高(P < 0.05;P < 0.05)。结论 通过多因素(饮食+气候环境+致病因子+局部用药+全身用药)复合造模方法可以复制出痛风湿热蕴结证病证结合模型,而且AQP2、HSP70两个指标在判断痛风湿热蕴结证型时有着重要作用,AQP2和HSP70含量变化可作为痛风湿热蕴结证模型建立成功与否的评价参考指标。

Abstract:Objective A multi-factor modeling approach was used to establish a damp-heat model first, and a modified Coderre method was used to model gout in order to replicate the the gout animal model with damp-heat syndrome, to explore the significance of the expression of aquaporin-2(AQP2) and heat shock protein70(HSP70) in the gout model with damp-heat syndrome, and to make further experimental validation for the evaluation of model establishment index and clinical drug efficacy determination index.Methods 40 healthy male SD rats were randomly divided into 4 groups, normal control group, gout model group, gout model with damp-heat syndrome group, damp-heat syndrome model group. Each group had 10 rats. Models were set up (the specific method was provided in full text) and the following general conditions were observed such as appearance, mental status, food and drink intake and weight changes. The inflammation index of the ankle joint, the gait condition of each group at 72 hours of modeling, blood uric acid, urine AQP2, plasma HSP70 and other indexes were measured.Results Urinary AQP2 levels in the 4 model groups were significantly different; Compared with the normal control group, urinary AQP2 was significantly higher in the gout model with damp-heat syndrome group and damp-heat syndrome model group (P < 0.05; P < 0.05); Urinary HSP70 levels in the 4 model groups were significantly different; Compared with the normal control group, plasma HSP70 was significantly higher in the gout model with damp-heat syndrome group and damp-heat syndrome model group (P < 0.05; P < 0.05); Compared with the gout model group, plasma HSP70 was significantly higher in the he gout model with damp-heat syndrome group and damp-heat syndrome model group (P < 0.05; P < 0.05).Conclusion Through the muiti factor diet and climate and pathogenic factor and topical and systemic medication combined modeling method can build the gout animal model with damp-heat syndrome. The AQP2 and the HSP70 index plays an important role in judging the painful rheumatism heat accumulation types. AQP2 and HSP70 content can be established for the pain of rheumatic fever stagnation syndrome model and evaluation index of success or not.
