毛细管电泳法测定罗布麻叶中芦丁、异槲皮苷和槲皮素的含量

摘    要:目的建立毛细管电泳法(capillary zone electrophoresis,CZE)同时分离测定罗布麻叶中的芦丁、异槲皮苷和槲皮素3种黄酮类化合物。方法采用熔融石英毛细管(58.5 cm×75μm),有效长度为48.5 cm(未涂层);缓冲体系为50mmol/L Na2B4O7(pH9.6+15%乙腈);分离电压20 kV(+)→(-);电泳时间20 min;电泳温度25℃;压力进样0.5 psi×20s;柱上检测UV 350 nm(二极管阵列检测器)。结果 3种黄酮类化合物在25 min内得到较好分离,芦丁、异槲皮苷、槲皮素的线性范围分别为4.4~442、.82~28.2、3.8~38μg/ml,相关系数为0.9968、0.99950、.9991,与峰面积线性关系良好;样品平均回收率分别为97.3%、98.4%和98.3%。结论本方法准确可靠,可用于罗布麻叶中黄酮类成分的含量测定。

Abstract:Objective To establish a high-performance capillary electrophoretic(HPCE) method for simultaneous determination of contents of rutin,isoquercitrin,and quercetin in Apocynum venetum.Methods A bare fused-silica capillary(58.5 cm×75 μm,effective length of 48.5 cm)was used.The buffer was composed of 50 mmol/L Na2B4O7(pH 9.60).The running voltage was set at 20 kV with polarity from anode(inlet) to cathode(outlet).The separation time was 20 min and temperature was set at 25 ℃.Online detection was performed at 350 nm(PDA).Results The three types of flavonoids were well separated in 25 minutes and linear relationship in the peak area was good.The linear range was 4.4-44,2.8-28.2,and 3.8-38 μg/ml,respectively.The correlation coefficient was 0.9968,0.9995,and 0.9991,respectively.The average recovery was 97.3%,98.4%,and 98.3%,respectively.Conclusions This method is simple,rapid,accurate,and reliable,which can be used for content determination of flavonoids in Apocynum leaves.
