血小板膜糖蛋白基因多态性与冠心病关系的研究

目的探讨我国汉族人群血小板膜糖蛋白(glycoprotein,GP)Ibα基因HPA-2多态性与冠心病的关系.方法选择403例确诊冠心病的患者和500例无心脏病史的健康体检者作研究对象,运用多聚酶链反应—限制性内切酶片段长度多态性技术(polymerase chain reaction-restriction fragment length polymorphism,PCR-RFLP)进行HPA-2多态性检测.结果所有年龄受试者中冠心病组Met/Met(M/M)+Thr/Met(T/M)基因型频率是13.2%,高于对照组的8.6%(P<0.05),比数比(odds ratio,OR)为1.6(95%可信区间1.1~2.4);年龄≤60岁的受试者中冠心病组T/M+M/M基因型频率(16.0%)同样高于对照组(8.8%)(P<0.05),OR值为2.0(95%可信区间1.1~2.4).结论血小板膜GPIbα中T/M和M/M基因型与冠心病之间存在相关性,在≤60岁的人中,此种相关性更强.

Association of platelet membrane glycoprotien gene polymorphism with coronary heart disease

Objective To investigate the correlation between the polymorphism of platelet membrane GPIbα gene and coronary heart disease(CHD).MethodsFour hunderd and three patients with CHD and 500 healthy controls without CHD were investigated.HPA-2 polymorphisms in GPIbα gene were checked by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP).ResultsThere were 3 genotypes: homozygous Thr/Thr(T/T) type,heterozygous Thr/Met(T/M) type and homozygous Met/Met(M/M) type.The prevalence of the M/M and T/M genotypes was higher in CHD group(13.2%) than that in the healthy controls group(8.6%)(P<0.05);odds ratio(OR) was 1.6(95%CI 1.1-4.3).In the subjects aged ≤60 years,the prevalence of the M/M and T/M genotypes in CHD group(16.0%) was also higher than that in the healthy controls(8.8%)(P<0.05);OR was 2.0(95%CI 1.1-2.4).ConclusionsThere is an correlation between the presence of the T/M and M/M genotypes in GPIbα gene and CHD.This correlation is stronger in patients aged ≤60 years.
