去甲肾上腺素预处理培养乳鼠心肌细胞丙二醛含量和细胞色素氧化酶活性观察

摘    要:目的 :对比观察去甲肾上腺素进行预处理 (norepinephrinepreconditioningNEPC)与经典缺血预处理 (is chemiapreconditioning ,IPC)对培养的乳鼠心肌细胞缺血 /再灌注损伤的影响 ,以探讨去甲肾上腺素预处理效应。方法 :应用培养的乳鼠心肌细胞缺血 /再灌注模型 ,将培养 5d的乳鼠心肌细胞随机分为 4组 :①正常对照组 ,②拟缺血再灌注组 (I/R) ,③缺血预处理组 (IPC) ,④去甲肾上腺素预处理组 (NEPC)。并分别进行细胞色素氧化酶 (CCO)活性测定和心肌细胞培养液中丙二醛 (MDA)测定。结果 :I/R组CCO酶活性降低 ,培养液中MDA含量增加 ,而IPC组和NEPC组CCO酶活性增高 ,MDA含量减少 ,均接近正常。结论 :去甲肾上腺素预处理和经典预处理均可取得类似的心肌保护的预处理效应

Abstract:Abstract: To investigate the effect of norepinepine preconditioning (NEPC) and classic ischemic preconditioning(IPC) on the ischemia/reperfusion(I/R) in cultured neonatal rat myocardium. Methods: The model of I/R in cultured neonatal rat myocardium was set up.After they had been cultured for 5 days,the cultured cardiac cells were divided into 4 groups:(1)The normal control group;(2)the I/R group;(3)the IPC group and (4)the NEPC group.They was compared by investigating the changes in MDA contents and CCO activities. Results: The result showed that as compared with the normal control group, the decrease on CCO activity and increase on the formation of MDA were the most serious in Group I/R, but is diminished greatly in Group NEPC and Group IPC. Conclusion: NEPC could reduce formation of MDA and improve CCO activeties. The result indicates that there is a similar protective effect on I/R injury of neonatal rat cardiomyocytes between NEPC and IPC.
