循环中内皮祖细胞与原发性高血压的关系

摘    要:目的探讨循环内皮祖细胞(EPCs)数量和功能与原发性高血压的关系。方法对34例初诊原发性高血压患者以及30例健康人采用密度梯度离心法获取外周血单个核细胞,将其接种在人纤维连接蛋白包被的培养板,培养7天后,收集贴壁细胞,多波长激光共聚焦显微镜鉴定FITC-UEA-Ⅰ和DiI-acLDL双染色阳性细胞为正在分化的EPCs,流式细胞仪检测其表面标志进一步鉴定EPCs,倒置荧光显微镜下计数。采用MTT比色法、改良的Boyden小室、粘附能力测定实验分别观察EPCs的增殖能力、迁移能力、粘附能力。结果原发性高血压患者循环中EPCs数量比治疗前明显减少(P<0.01),降压达标后原发性高血压患者EPCs数量增加,但与对照组相比,达标后原发性高血压患者EPCs数量仍然少(P<0.05)。原发性高血压患者EPCs的增殖、迁移、贴壁能力受损(P均<0.01);降压达标后的原发性高血压患者EPCs的增殖、迁移、贴壁能力受损没有完全恢复(P均<0.01)。结论原发性高血压患者循环EPCs数量减少,功能受损;血压增高是循环EPCs数量和功能改变的原因之一,循环EPCs数量和功能的改变可能还参与原发性高血压的发生和发展。

Abstract:Objective To study the relationship between circulating endothelial progenitor cells(EPCs) and essential hypertension.Methods 34 patients with primary diagnosed as essential hypertension and 30 healthy individuals were involved in this study.Mononuclear cells were isolated by density gradient centrifugation.EPCs were characterized as adherent cells double positive staining for DiI-acLDL uptake and lectin binding with using fluorescent microscope,EPCs proliferation and migration were assayed by MTT assay and modified Boyden chamber assay,respectively,EPCs adhesion assay was performed by replating cells on fibronectin-coated dishes,then counting the adhesion cells with inverse microscope.Results The number of circulating EPCs in patients with essential hypertension was significantly reduced in comparison to treatment before(P<0.01),an elevated EPCs were obtained in patients after the blood pressure was controlled.While compared to control group,the number of EPCs was still lower(P<0.05).In addition,EPCs proliferative,migratory and adhesive capacity in patients with essential hypertension show senescence(P<0.01) and they recovered to some degree after the patients blood pressure were controlled(P<0.01).Conclusion The present findings suggested that circulating EPCs be impaired in essential hypertension patients,and circulating EPCs may be involved in the mechanism of essential hypertension.
