摘    要:目的研究sorcin基因与白血病多药耐药的相关性。方法用NorthernBlot和WesternBlot方法,检测人白血病耐药细胞株K562/A02及其野生型细胞株K562中sorcin基因和蛋白表达水平的差异。设计针对sorcin基因的反义寡核苷酸,通过脂质体将其导入K562/A02细胞,通过WesternBlot方法检测转染前后细胞内sorcin蛋白表达的变化,运用MTT方法检测转染反义寡核苷酸后细胞对阿霉素敏感性的变化。结果NorthernBlot和WesternBlot结果表明,K562/A02细胞中sorcin表达水平明显高于K562敏感细胞。将sorcin基因反义寡核苷酸转入K562/A02细胞后,细胞内sorcin蛋白表达明显下降,对阿霉素的敏感性提高。结论作为一种耐药相关基因,sorcin参与了白血病多药耐药细胞耐药表型的形成,有望成为今后白血病耐药诊断和治疗的新靶点。

Abstract:Aim To explore the role of sorcin gene on the multidrug resistance phenotype of human leukemia cell line K562/A02. Methods The difference of sorcin expression level between K562/A02 and its parental cell line K562 were determined using Northern blot and Western blot. An antisense oligodeoxynucleotide (ASODN) targeted sorcin was transfected to K562/A02 using Oligofectamine. The expression of sorcin was measured using Western blot. The sensitivity of cells to doxorubicin was measured using MTT assay. Results The sorcin level was significantly higher in K562/A02 cells than in K562 cells . Transfection of K562/A02 cells with ASODN targeted to sorcin reduced the level of sorcin expression and increased the sensitivity of K562/A02 cell to doxorubicin. Conclusion Sorcin, an important gene involved in MDR in leukemia cells, may be a potential target for MDR modulators investigation in the future.
