摘    要:目的 研究人鼻咽癌细胞形态、DNA含量及倍体与放射敏感性的异质性之间的关系 ,探讨预测肿瘤放射敏感性的可行方法。方法 应用 2种放射敏感性不同的鼻咽癌细胞系CNE 2Z亚克隆株F1、S1,通过细胞培养及裸鼠体内实验 ,用HE染色法、Feulgen染色法和图像分析仪 ,观察这 2株细胞及其裸鼠移植瘤的形态学参数、DNA含量及倍体分布。 结果 形态参数测定表明 ,F1细胞面积、细胞体积等参数小于S1(P <0 .0 1) ,核周长、核直径大于S1(P <0 .0 1) ,核质比大于S1(P <0 .0 1)。F1细胞及裸鼠移植瘤DNA含量、DI值、5C及 >5C比例均高于S1组 (P <0 .0 1)。结论 F1、S1放射敏感性的异质性与细胞分化程度、DNA含量及 >5C比例有关 ,采用计算机图像分析法定量分析 ,可以预测鼻咽癌细胞间的放射敏感性差异

Abstract:Objective To investigate the relationship of radiosensitive heterogeneity and cellular morphology,DNA content and ploidy of human nasopharyngeal carcinoma cells,and to explore the possible methods for estimating radiosensitivity.Methods Two subclones of human nasopharyngeal carcinoma cell line CNE-2Z:F1 and S1 with different radiosensitivity,were used for cell culture and nude mice experiment in vivo.Two cell strains and transplanted tumors were assessd for different morphological parametes,DNA content and ploidy distribution with HE staining,Feulgen staining and image analysis.Results Morphological paramete detection were showed that parametes of F1 were lower than those of S1 in cell area,cell size(P<0.01).Peri-length and diameter of nucleus of F1 were higher than those of S1(P<0.01).The nuclear/cytoplasm ratio of F1 was higher than that of S1(P<0.01).DNA content,DI value,5C and >5C of F1 and transplanted tumors were higher than those of S1 group with a great statistical significance(P<0.01).Conclusion The results suggest that cell differentiation,DNA content and >5C be correlated with radiosensitive heterogeneity of F1 and S1.It is indicated that quantitative estimation with computer image analysis holds possibility in predicting the radiosensitivity of nasopharyngeal carcinoma cells.
