摘    要:目的:研究血必净注射液及其含有的10种化合物对P815细胞脱颗粒、组胺和氨基己糖苷酶释放的影响。方法:选取P815细胞作为体外过敏模型,通过CCK-8法测定各组分的IC10作为给药浓度,C48/80作为阳性药,采用中性红染色,计算出各组脱颗粒百分率;采用ELISA法测定各组分上清液中组胺的释放量;采用底物法检测各组上清液中氨基己糖苷酶的释放度。结果:与空白组相比,绿原酸、蒿本内酯、咖啡酸、丹酚酸B和血必净组细胞的脱颗粒百分率存在极显著差异;绿原酸、咖啡酸对组胺释放量的影响最大,且显著增加细胞中的氨基己糖苷酶释放量。结论:血必净注射液会刺激P815细胞释放过敏相关物质,该反应很可能与其含有的绿原酸、咖啡酸、原儿茶醛、蒿本内酯和丹酚酸B等成分有关。

Abstract:Objective: To study the effects of Xuebijing injections and their 10 compounds on degranulation, histamine and hexosaminidase release of P815 cells. Methods: P815 cells were selected as the allergic model in vitro. IC10 of each component was determined by a CCK-8 method for dosing, and C48/80 was used as the positive drug. The degranulation percentage of each group was calculated by neutral red staining. The release of histamine in the supernatant was determined by ELISA. The release of aminohexosidase in the supernatant was detected by a substrate method. Results: Compared with the blank group, the percentages of degranulation of cells were significantly different in the chlorogenic acid, artemisinolide, caffeic acid, salvianolic acid B and Xuebijing injection groups; chlorogenic acid and caffeic acid had the greatest effects on the release of histamine and hexosaminidase in cells. Conclusion: Xuebijing injections stimulate P815 cells to release allergy-related substances, and the reactions are most likely related to their components chlorogenic acid, caffeic acid, protocatechualdehyde, artediolide and salvianolic acid B.
