基于网络药理学探讨桂芍配伍治疗骨关节炎的作用机制

目的运用网络药理学技术研究“桂枝—白芍”药对治疗骨关节炎的主要活性成分和靶点,进而探讨桂芍治疗骨关节炎的潜在作用机制。方法通过检测网络数据库平台(TTD、DrugBank和GeneCards)数据库构建骨关节炎疾病靶点;通过检测TCMSP、HIT数据库筛选桂枝—白芍药对活性成分及靶点。利用Cytoscape3.7.2软件构建活性成分—靶点网络图,并对靶点进行KEGG及GO富集分析。结果从“桂枝—白芍”药对中共筛选出17个有效成分,与骨关节炎相关作用靶点40个,KEGG与GO富集分析显示“桂枝—白芍”药对主要与癌症、免疫、炎症以及细胞衰老、凋亡等信号通路有关。结论本研究利用网络药理学的方法,初步揭示了“桂枝—白芍”药对对于骨关节炎的主要活性成分、治疗靶点及相关信号通路,为进一步深入研究其作用机制提供了参考。

Exploration on action mechanism of the combination of Ramulus Cinnamoni and Radix Paeoniae Alba in the treatment of osteoarthritis based on network pharmacology

Objective To study the main active ingredients and targets of medicine pair of‘Ramulus Cinnamoni—Radix Paeoniae Alba’in the treatment of osteoarthritis by using network pharmacology technology,so as to explore the potential action mechanism.Methods Osteoarthritis disease’targets were established by testing the database on database platforms,including TTD,DrugBank and GeneCards.The active components and targets of the medicine pair of‘Ramulus Cinnamoni—Radix Paeoniae Alba’were screened by detecting TCMSP and HIT database.Cytoscape3.7.2 software was used to construct the active component-target network diagram,and KEGG and GO enrichment analysis was performed on the targets.Results A total of 17 active components were screened from the medicine pair of‘R amulus Cinnamoni—Radix Paeoniae Alba’,and 40 target genes related to osteoarthritis were identified.KEGG and GO enrichment analysis showed that the medicine pair of‘R amulus Cinnamoni—Radix Paeoniae Alba’were mainly associated with cancer,immunity,inflammation and cell senescence,apoptosis and other signaling pathways.Conclusion In this study,using the method of network pharmacology,the main active components,therapeutic targets and related signal pathways of the medicine pair of‘R amulus Cinnamoni—Radix Paeoniae Alba’in the treatment of osteoarthritis,were preliminarily revealed.It provides a reference for further research on its mechanism of action.
