IGF2在先兆子痫产妇胎盘中无印迹丢失

【目的】通过检测印迹基因IGF2在先兆子痫产妇胎盘中的特殊等位基因的表达,探讨IGF2印迹状态与先兆子痫发病的关系。【方法】收集经剖宫产的正常足月妊娠及先兆子痫产妇胎盘,从胎盘中提取基因组DNA,经PCR扩增、Apa1酶切筛选杂合子。提取杂合子胎盘中的总RNA并逆转录合成cDNA,cDNA经PCR扩增、内切酶消化,以评价IGF2特殊等位基因的表达。【结果】IGF2在正常足月胎盘中呈印迹状态;在先兆子痫胎盘中,IGF2为单等位基因表达,无杂合子状态。【结论】在先兆子痫胎盘中未发生IGF2的印迹丢失。

Maintenance of normal imprinting of insulin-like growth factor-2 gene in placenta of preeclampsia

Objective] To investigate the hypothesis of abnormal expression of the imprinted IGF2 gene in relation to the etiology of preeclampsia.Methods]Normal and preeclamptic placental tissues were collected during cesarean delivery,genomic DNA from placenta was amplified by polymerase chain rection(PCR)and screened for heterozygosity at IGF2 with Apa1.If heterozygous presented,polyadenylated RNA was isolated and reverse transcribed into cDNA.cDNA then was amplified by PCR and the products were digested with restriction enzymes to evaluated the expression of IGF2.Results]IGF2 was imprinted in full-term placenta.No loss of imprinting,as determined by the finding of biallelic expression,and null-mutation was detected in any case of preeclampsia.Conclusions]Our data suggest that IGF2 gene maintains its imprinting in placenta from preeclamptic patients.
