摘    要:目的研究慢性阻塞性肺疾病(COPD)患者和脂多糖(LPS)刺激后健康者中痰巨噬细胞趋化因子的表达。方法3组患者运用最小限度侵入法获得诱导痰,纯化后得到小巨噬细胞,对单核细胞趋化蛋白作定量测定。结果COPD组和LPS组的痰液小巨噬细胞水平与对照组相比明显增加,LPS组痰液小巨噬细胞水平与吸入LPS前相比明显增加。C()PD患者小巨噬细胞和单核细胞趋化蛋白CCL2、CCL7、CCL13和CCL22的mRNA水平增加,其中CCL13mRNA水平增加100多倍。LPS诱导痰液小巨噬细胞趋化因子表达模式中,CCL2、CCL7向上调节,CCL13向下调节。结论与LPS诱导痰液小巨噬细胞不同,COPD中痰液小巨噬细胞可以主动诱导一系列CCL趋化因子。

Gene Expression of Monocyte Chemoattractant Protein CCL in Sputum Macrophages of Patients with Chronic Obstructive Pulmonary Disease

Abstract:Objective To study the expression of chemokines in sputum macrophages of patients with chronic obstructive pulmonary disease (COPD) and lipolysaccharide (LPS)-stimulated healthy people. Methods By the minimally invasive method, the sputum induction in three groups was obtained and purified to get macrophages, and monocyte chemoattractant proteins were quantitatively determined. Results In COPD and LPS groups, the levels of small sputum macrophages were obviously increased compared with control group. Small sputum macrophages were increased significantly after LPS inhalation. The levels of monocyte chemoattractant proteins CCL2, CCL7, CCIA3, CCL22 were increased in COPD group, in which CCL13 mRNA was increased by 100 times. The expression mode of LPS-induced chemokines was different from COPD in that CCL2 and CCL7 were increased, while CCL13 level in small macrophages was decreased. Conclusion Compared with LPS-induced sputum macrophages, the sputum macrophages in COPD can spontaneously induce a series of CCL chemokines, with marked increases in CCL13.
