葛根异黄酮对大鼠前列腺增生的影响

摘    要:目的 探讨葛根异黄酮对丙酸睾酮诱导的去势大鼠前列腺增生的抑制作用及其发生机制.方法 雄性Wistar大鼠,随机分为6组.空白组大鼠行假手术(其余均行去势手术).饲养7 d后,除空白组注射生理盐水外,其余5组均皮下注射丙酸睾酮10 mg/(kg·d),连续注射10 d,复制前列腺增生模型.随后隔两日一次皮下注射丙酸睾酮维持,阳性对照组灌胃非那雄胺1.0 mg/(kg·d),葛根异黄酮高、中、低剂量组按照180、120、60 mg/(kg·d)灌胃,其余两组给予生理盐水,连续给药35 d.取血清,测定前列腺特异性酸性磷酸酶活力;分离前列腺组织,称重,计算前列腺指数(PI);用足趾容积测量仪测前列腺体积的变化;光镜下观察前列腺组织形态的变化.结果 各剂量组葛根异黄酮对大鼠前列腺湿重、体积及PI相对模型组有明显影响(P〈0.05).组织形态学观察,不同剂量的葛根组均可缓解由丙酸睾酮所致的大鼠前列腺增生的症状,表现为上皮变薄,腺腔内分泌物减少,间质减少等.结论 葛根异黄酮对大鼠前列腺增生有一定抑制作用.

The influence of puerariae isoflavones on benign prostatic hyperplasia in emasculated rat

Abstract:Objective To study the inhibitory effect of pueraria isoflavones on prostate hyperplasia induced by testos- terone in emasculated rats and clarify its mechanism. Methods 72 male Wistar rats were randomly divided into 6 groups, the control group ( only Normal Saline, NS ) , the finasteride group [ 1.0 mg/( kg · d) ] , the model group, the pueraria isoflavones group 1180,120,60mg/ (kg · d) respectively 1. The rats in the control group underwent sham op- eration, the rest were emasculated operation. From the 8th day, except the blank control group, rats in the above-men- tioned 5 groups were given Testosterone Propionate [ 10 mg/( kg · d) ] by subcutaneous injection for continuous 10 days to copy prostatic hyperplasia model. Then maintaining with subcutaneous injection once every two days, the blank control group and model group were given normal saline [ 1.0 mg/( kg · d) ] , the finasteride group was given finas- teride · 1.0 mg/(kg · d) 1 ,the pueraria isoflavones groups were given for 35 days. On the 36th day,serum was taken and the PAP vitality was determined. Then the prostate was separated, and weighed, the index was calculated, prostate volume change using the toe volumetric measuring instrument and the histology and morphology of the prostate was oh- served. Results Pueraria isoflavone in different dose groups had significant effect on wet weight, volume and index of rat prostate ( P 〈 0.05 ). And alleviated the change of prostate caused by Testosterone Propionate : epithelium thinning,luminal secretion and interstitial substance decreased. Conclusion Pueraria isoflavone had inhibitory effect on benign prostatic hyperplasia in rats.
