摘    要:目的 研究去甲斑蝥素(NCTD)对白血病细胞死亡受体(DR)5表达的影响.方法不同浓度NCTD处理K562和HL-60细胞16 h后,流式细胞术分析DR5表达和细胞内活性氧(ROS)相对含量;通过免疫印迹技术分析激酶JNK1的表达.结果 NCTD以浓度依赖方式上调DR5表达,细胞内活性氧含量和JNK1表达也随NCTD浓度增大而增加.结论 NCTD可能通过ROS/JNK途径上调DR5表达.

Abstract:Objective To explore the expression of DR5 on leukemia cells induced by NCTD. Methods Leukemia cells K562 and HL-60 were cultured in vitro and treated by different concentration of NCTD for 16 hours. The expres- sion of DR5 and ROS in cells were measured by flow cytometry, and the content of JNK1 in cells was detected by west- ernblot assay. Results It was found that NCTD could up-regulated DR5 on leukemia cells in a dose-dependent man- ner. Both the levels of ROS and JNK1 rise with increasing in the concentration of NCTD. Conclusion The expres- sion of DR5 on leukemia cells could be up-regulated by NCTD through ROS/JNK signaling pathway.
