摘    要:目的 研究雷帕霉素靶蛋白(mammalian target of rapamycin,mTOR)、Akt蛋白在食管鳞状细胞癌(ESCC)及周围正常黏膜组织中差异表达状况,探讨两者间相互关系及在ESCC中的作用.方法 利用免疫组织化学染色技术研究mTOR与Akt蛋白在94例正常食管黏膜组织及ESCC石蜡包埋组织中的表达情况,分析其临床病理意义.结果 与相应正常食管黏膜组织相比,ESCC组织中mTOR和Akt蛋白表达明显上调.在正常食管黏膜组织及ESCC组织中,mTOR蛋白的阳性率分别是28.6%和52.1%,差别具有统计学意义((χ2=10.175,P〈0.05);Akt的阳性率分别为20.2%及42.6%,差别具有统计学意义((χ2=10.148,P〈0.05).结论 食管鳞状细胞癌与mTOR及Akt蛋白的表达有密切相关性.

Abstract:Objective To investigate the differential expression of mTOR and Akt protein in malignant and normal esophageal tissues, elucidate its correlation to some clinicopathologic parameters of esophageal squamous cell carcinoma, and evaluate its clinic significance. Methods The expression of mTOR,Akt protein in 94 specimens of esophageal squamous cell carcinoma tissues and corresponding noncan- cerous tissues were detected by immunohistochemistry using the streptaridinperoxidase method. The relationship between mTOR, Akt protein and pathology of esophageal squamous cell carcinoma was further demonstrated. Results The expression of mTOR and AKT proteins was sig- nificantly higher in cancerous tissue specimens than that of the normal tissues ( P 〈 0.05 ). Specifically, roTOR protein was expressed in 28.6% and 52. 1% cases of normal esophageal mucosa and malignant esophageal tissues,respectively,while the expression of AKT protein occurred in 20.2% and 42.6% cases of normal esophageal mucosa and malignant esophageal tissues, respectively. The increased expression of these two proteins between normal and malignant esophageal tissues had statistically significant difference (χ2 = 10. 175 ,P 〈 0.05 andx2 - 10. 148 ,P 〈 0.05, respectively). Conclusion ESCC is closely related to the expression of roTOR and Akt protein.
