糖代谢异常对冠心病相关血清微小RNA表达的影响

摘    要:目的检测相关血清微小RNA(miRNA)在冠心病及糖尿病合并冠心病患者中的表达水平,探讨糖代谢异常对冠心病相关血清miR-126、miR-146表达的影响。方法选择患者48例,分别为对照组15例,冠心病组17例,糖尿病合并冠心病组(合并组)16例。采用TaqMan探针实时定量PCR技术,检测各组血清miR-126、miR-146;采用TaqMan实时荧光定量PCR对血清中靶miRNA丰度进行定量。结果与对照组比较,冠心病组及合并组患者miR-126基因表达明显上调(P<0.05),而miR-146基因表达虽有下调趋势,但差异无统计学意义(P>0.05)。Spearman相关分析显示,CT miR-126与空腹血糖、餐后2h血糖水平呈负相关(r=-0.686,P=0.002;r=-0.723,P=0.001)。结论糖代谢异常对冠心病相关血清miR-126的表达产生影响,从而在一定程度上为miR-126成为糖尿病大血管病变的血清学检测指标提供理论依据。

Effect of abnormal glucose metabolism on expression of serum microRNA in patients with coronary heart disease
Abstract:Objective To study the effect of abnormal glucose metabolism on expression of serum microRNAs(miR-126,miR-146)in patients with diabetes mellitus(DM) accompanying coronary heart disease(CHD).Methods Forty-eight patients were divided into control group(n=15),CHD group(n=17)and DM accompanying CHD group(n=16).Their serum miR-126 and miR-146 levels were measured by quantitative RT-PCR with TaqMan probes and their serum target miRNA fraction was quantified by fluorescence RT-PCR with TaqMan probes.Results The expression level of miR-126 was significantly higher in CHD group and DM accompanying CHD group than in control group(P<0.05).No significant difference was found in expression level of miR-146 among the 3 groups although it was lower in CHD group and DM accompanying CHD group than in control group(P>0.05).Spearman correlation analysis showed that the cycle threshold(CT)of miR-126 was negatively correlated with the fasting plasma glucose level(r=-0.686,P=0.002)and 2 h postprandial glucose level(r=-0.723,P=0.001).Conclusion Abnormal glucose metabolism influences the expression of miR-126 in CHD patients and can thus provide the evidence for detectiing large blood vessel lesions in DM patients.
