家蝇幼虫血淋巴纯化蛋白对Hela肿瘤细胞抑制作用的形态学观察

摘    要:目的 研究家蝇幼虫血淋巴纯化蛋白(anticancer protein from Musca domestica larva hemolymph,MAC-1)对体外培养的人官颈癌Hela细胞抑制作用引起的形态学改变.方法 应用四甲基偶氮噻唑蓝(MTT)法、光学显微镜、HE和吖啶橙(AO)染色、透射电镜(SEM)的方法检测MAC-1对体外培养的人宫颈癌Hela细胞的抑制作用及形态学改变.结果 MAC-1对人宫颈癌Hela细胞株的增殖抑制作用呈剂量依赖性,同时呈时间依赖性.MAC-1 12.5 μg/ml作用12h后在倒置显微镜、HE及AO染色和透射电镜均观察到典型细胞凋亡形态变化.结论 MAC-1对人宫颈癌Hela细胞株具有抑制增殖及诱导凋亡作用.
