奥沙利铂长循环脂质体对人结直肠癌SW480细胞凋亡影响

摘    要:目的探讨奥沙利铂长循环脂质体对人结直肠癌SW480细胞的凋亡作用。方法采用荧光标记的长循环脂质体,奥沙利铂长循环脂质体和游离奥沙利铂分别处理人结直肠癌SW480细胞,流式细胞仪、荧光显微镜检测细胞对脂质体的摄取;MTT分析载药脂质体对细胞增殖影响;透射电镜观察细胞超微结构变化;流式细胞仪及TUNEL检测细胞凋亡;Western blot分析actived-caspase-3(P17)的表达。结果脂质体被细胞摄取,2 h的荧光强度为(197.72±8.92),24 h的荧光强度为(641.74±44.46);2.6 mmol/L的奥沙利铂长循环脂质体(含28 mg/L奥沙利铂)对细胞的活性影响及凋亡诱导作用大于28 mg/L游离奥沙利铂(P<0.01);actived-caspase-3蛋白的表达随细胞凋亡增加而上调(P<0.01)。结论奥沙利铂长循环脂质体对人结直肠癌SW480细胞有显著凋亡诱导作用。
