| 摘 要： | 目的探讨蛋白质二硫键异构酶A3(protein disulfide isomerase A3,PDIA3)在大鼠同种异体移植肾组织中的表达及意义。方法实验分2组。实验组(异基因移植组):近交系雄性F344和Lewis大鼠分别作为供体和受体,共8对。对照组(同基因移植组):近交系雄性F344大鼠作为供体和受体,共8对。分别建立原位大鼠肾移植模型。术后7 d获取移植肾组织,分别应用免疫组化、RT-PCR和Western blot等方法检测实验组和对照组移植肾组织中的PDIA3的表达情况。结果 PDIA3主要表达在细胞质,在实验组移植肾组织中的表达呈阳性或强阳性,而在对照组表达呈弱阳性。实验组移植肾PDIA3的mRNA表达量为(0.821±0.046),对照组移植肾PDIA3的mRNA表达量为(0.187±0.154),2组比较差异有统计学意义(P<0.01)。实验组移植肾PDIA3的蛋白表达量为(0.812±0.045),对照组移植肾PDIA3的蛋白表达量为(0.237±0.134),2组比较差异有统计学意义(P<0.01)。结论大鼠同种异基因肾移植急性排斥反应组移植肾组织中的PDIA3的表达明显高于同基因移植对照组。PDIA3可能参与肾移植急性排斥反应的发生、发展过程。

| Abstract: | Objective To investigate the expressions of protein disulfide isomerase A3(PDIA3) in renal allograft of rats.Methods The rat model of allogene renal transplantation was established in the 2 groups,homogenic transplantation group(F344→F344,8 pairs) and allogene transplantation group(F344→Lewis,8 pairs).In 7 d after the transfection,the graft was taken out.The expression of PDIA3 in the kidney was detected by immunohistochemistry,RT-PCR and Western blot analysis.Results Immunohistochemistry indicated that the expression of PDIA3 was increased significantly in the allogene transplantation group than in the homogenic transplantation group.Compared with control group,the mRNA expression as well as protein expression of PDIA3 was increased significantly in the allogene transplantation group than in the control(P<0.01).Conclusion PDIA3 is highly expressed in the allogene transplantation rats than in the homogenic transplantation rats,and may play an important role in acute renal allograft rejection. |
