摘    要:目的: 分析银线莲水提物中的胞苷、尿苷、腺嘌呤、肌苷、鸟苷、腺苷6种核苷类物质。方法: 高效液相色谱法-电喷雾电离/离子阱质谱法(HPLC-ESI/MS)对银线莲中超声波辅助萃取法水提物中6种核苷类物质进行测定和鉴别。结果: 银线莲药材中检测到胞苷、尿苷、腺嘌呤、肌苷、鸟苷和腺苷6种核苷成分。6种核苷类物质质量浓度在1.25~80.0 μg·ml-1范围内呈良好线性关系(r≥0.999 4),回收率在92.7%~96.8%之间,RSD≤1.6%。测得银线莲药材提取物中胞苷、尿苷、腺嘌呤、肌苷、鸟苷和腺苷含量分别为0.106 2,0.339 1,0.069 9,0.082 0,0.030 8,0.107 4 mg·g-1结论: 此法简便、灵敏快速,首次报道银线莲中含有重要药理活性的6种核苷类物质,为银线莲的药理和临床试验研究提供理论基础。

Abstract:OBJECTIVE To analyze the six nucleosides (cytidine, uridine, adenine, inosine, guanosine and adenosine) from aqueous extract of Goodyera matsumurana. METHODS The six nucleosides were extracted from Goodyera matsumurana by ultrasonic-assisted aqueous extraction, and were identified and determined by using high performance liquid chromatography-electrospray ionization/ion traps mass spectrometry (HPLC-ESI/MS). RESULTS Cytidine, uridine, adenine, inosine, guanosine and adenosine in Goodyera matsumurana were identified. The linearity was good within the concentration range of 1.25-80 μg·ml-1 (r≥0.999 4) for the six nucleosides.The average recovery of them were 92.7%-96.8% with RSD≤1.6%. The content of six nucleosides (cytidine, uridine, adenine, inosine, guanosine and adenosine) from Goodyera matsumurana were 0.106 2 mg·g-1, 0.339 1 mg·g-1, 0.069 9 mg·g-1, 0.082 0 mg·g-1,0.030 8 mg·g-1 and 0.107 4 mg·g-1, respectively. CONCLUSION The proposed method is simple, accurate and rapid. So far there is no report indicating that Goodyera matsumurana contains these six important bioactive nucleoside compounds. It provides a scientific basis for further pharmacological and clinical studies on Goodyera matsumurana.
