IL-10联合生长抑素对大鼠重症急性胰腺炎炎症相关细胞因子的抑制作用

摘    要:目的探讨白细胞介素10(IL-10)联合生长抑素对大鼠重症急性胰腺炎(SAP)炎性因子的调节作用。方法将27只SD大鼠依据随机数字表法分为三组:A组为空白对照组(n=6),B组为SAP组(n=6),C组为生长抑素联合IL-10肌肉注射治疗组(n=15)。其中B组采用"逆行胆胰管注射法"制成SAP模型,C组在模型建立后即刻注射IL-10和生长抑素。三组均于术后处死大鼠且于第1、6、12小时留取血液标本,采用酶联免疫吸附法测定血清中炎性因子肿瘤坏死因子α(TNF-α)、IL-6、IL-8的变化。结果 B组和C组第6小时、第12小时的IL-6、IL-8、TNF-α水平均有升高,且呈上升趋势,差异有统计学意义(P<0.05),但C组较B组IL-6、IL-8、TNF-α水平低,差异有统计学意义(P<0.05)。结论 SAP时,应用IL-10联合生长抑素可降低血中IL-6、IL-8、TNF-α的水平,是具有临床应用前景的治疗SAP的方法之一。

Abstract:Objective To investigate the effects of interleukin 10 (IL-10) combined with somatostatin treatment on theinflammatory factors in severe acute panereatitis(SAP) rat model. Methods Twenty-seven SD rats were randomly divided into three groups : group A was control group ( n = 6 ), group B was SAP group (n = 6 ) , group C was somatostatin intramuscular injection in combination with IL-10 intramuscular injection treatment group( n = 15 ). Group B was made into SAP models by retrograde eholangiopanereatography injection method. Group C was immediately injected with IL-10 and somatostatin. Serum were collected from the groups 1 hour ,6 hours, 12 hours after treatment, ELISA assay of serum inflammatory factors ( TNF-ct, L-6, IL-8 ) were performed. Results Increase of IL-6, IL-8, TNF-αlevels at 6 h, 12 h were observed in both group B and group C, and a rising trend was observed, the difference was statistically significant ( P 〈 0.05 ), but IL-6, IL-8,TNF-α level of group C were lower than group B, the difference was statistically significant ( P 〈 0.05 ). Conclusion In SAP model, the application of IL-10 combined with somatostatin, could significantly inhibit serum levels of IL-6, IL-8, TNF-α, which is one of the treatments with perspectives for the clinical application.
