摘    要:TET1作为TET蛋白家族的一员,是一种α-酮戊二酸和Fe2+依赖的双加氧酶,能转化5-甲基胞嘧啶(5m C)为5-羟甲基胞嘧啶(5hm C),从而启动DNA去甲基化程序。TET1既能对胚胎干细胞分化基因保持沉默,又能使其多能性基因保持激活,这种双管齐下的调控方式保持了胚胎干细胞自我更新能力和多能性潜力。TET1与肿瘤、精神疾病及白血病等疾病密切相关,因此TET1的研究对于拓展DNA去甲基化、胚胎干细胞基因调控及探求临床疾病治疗新靶向具有潜在价值。

Research Progress of DNA Methylation Hydroxylase TET1

Abstract:TET( ten eleven translocation) 1 is a member of the TETproteins family,which is anα-ketoglutarate and Fe2 +-dependent dioxygenase,and can convert 5-methylcytosine( 5m C) to 5-hydroxymethylcytosine( 5hm C),and start DNA demethylation process. TET1 can keep not only differentiation gene of embryonic stem cells remaining silent but also pluripotent gene remaining activated,and this twopronged regulation approach can maintain the embryonic stem cells self-renewal and pluripotency potential. Furthermore,TET1 is closely associated with cancer,mental disorders,leukemia and other diseases.Therefore,the investigation on TET1 has a certain value for expanding understanding of DNA demethylation,regulating embryonic stem cells genes and exploring a new targeted therapy for the clinical diseases.
