| 摘 要： | 本文介绍用McAb作为包被和检测抗体进行双抗体夹心ELISA来测定人血浆纤维连结蛋白。通过实验找出最佳条件,即两种McAb混合使用,在McAb和酶结合物稀释液内加入4％PEG及使用TMBI底物的最适浓度等,该法敏感性可达80ng/ml,可作为临床常规方法使用。

| Abstract: | A solid-phase enzyme immunoassay was developed for the quantitation of fi-bronectin. Two monoclonal antibodies against human plasma fibronectin and the different combination of them were compared for optimal conditions in a double antibody sandwich ELISA system. It was found that immunological response of mixture of McAb is better than that of single Me Ab. Sensitivity of the test w.as increased by adding 4%PEG to incubation buffers of the detector antibody and the conjugate and using a new substrate TMB in this system. The detection limit of the assay was 80ng/ml.It was sugested that the method wight be used for the quantitation of fibronectin in human plasma as a clinical routine one. |
