Abstract:Objective To explore the effects of lidocaine on the proliferation and invasion of ovarian cancer cells and related molecular mechanisms.Methods SKOV-3 cells in logarithmic growth phase were divided into 3 groups-control group,low concentration group and high concentration group according to the random number table method.The low-concentration group and the high-concentration group were treated with the final concentration of 0.1 mmol/L and 1.0 mmol/L lidocaine,and the control group were cultured with complete culture medium only.At 24 h and 36 h after treatment,CCK-8 method were used to detect cell proliferation rate,Transwell chamber method were used to detect cell migration and invasion,double staining method were used to detect cell apoptosis rate,and real-time fluorescent quantitative RT-PCR were used to detect phosphatidylinositol 3-kinase(PI3K)mRNA expression,Western blotting detection of PI3K protein expression.Results At 24 h and 36 h after treatment,the cell proliferation rate,the number of migrated cells,the number of invaded cells,and the relative expression levels of PI3K mRNA and protein of the low concentration group and the high concentration group were lower than those of the control group,and the high concentration group were lower than the low concentration group,compared the differences were statistically significant(P<0.05).The apoptosis rate in the low concentration group and the high concentration group were lower than those of the control group,and the high concentration group were lower than the low concentration group,compared the difference were statistically significant(P<0.05).Conclusion The application of lidocaine in ovarian cancer is beneficial to inhibit cell proliferation,migration and invasion,and promote cell apoptosis.Its molecular mechanism may be related to the inhibition of PI3K expression,and the effect of high concentration lidocaine in this study is more significant.
