Effect of Panax notoginseng saponins on neovascularization and expression of adiponectin,tumor necrosis factor-alpha of retinopathy of prematurity mice induced by hyperoxia

Abstract:Objective To investigate the effect of Panax notoginseng saponins on neovascularization and expression of adiponectin(APN),tumor necrosis factor-alpha(TNF-α)of retinopathy of prematurity(ROP)mice induced by hyperoxia.Methods Fourty-five mice were randomly divided into normal control group,ROP group and Panax notoginseng saponins,with 15 mice in each group,the ROP model was established by Smith menthod.Mice in the Panax notoginsenoside group were injected intraperitoneally with 4μL of Panax notoginseng saponin solution daily,and the normal control group and ROP group were injected intraperitoneally with the same dose of 0.9%sodium chloride solution daily.Heart perfusion was conducted and retinal flatmount was made to observe the retinal vascular morphology.The retinal endothelial cell nuclei was observed by HE staining.The expression levels of serum APN and TNF-αwere detected by enzyme-linked immunosorbent assay(ELISA).Results The retinal flatmount results showed that the vascular proliferation of ROP group was obvious,while the vascular proliferation of Panax notoginseng saponins group was decreased.HE results showed that the number of endothelial cells in the inner limiting membrane were massive,while the Panax notoginseng saponins group was significantly decreased.The number of endothelial cell nuclei in the ROP group and the panax notoginsenoside group were(24.20±6.26),(3.40±1.03),which were significantly higher than those of the normal control group(0.40±0.12),while the panax notoginsenoside group was significantly lower than the ROP group,and the difference was statistically significant(P<0.05).Compared with the normal control group(16.38±3.56)ng/L,(13.18±1.81)mg/L],the expression of serum TNF-αin the ROP group(33.23±6.96)ng/L,(7.23±1.33)mg/L]and the heptasaponin group(18.54±5.85)ng/L,(11.95±1.79)mg/L]was significantly increased,and the expression of APN was significantly decreased,and the differences were statistically significant(P<0.05).Compared with the ROP group,the expression of serum TNF-αin the panax notoginsenoside group was lower,and the expression of APN increased,and the differences were statistically significant(P<0.05).Conclusion Panax notoginseng saponins could siginificantly inhibit neovascularization,and the probable mechanism was up-regulate the expression level of APN and down-regulate the expression level of TNF-α.
