系统性红斑狼疮患者B淋巴细胞中EB病毒感染与bc1-2表达的相关研究

摘    要:目的 了解EB病毒、bc1 2、系统性红斑狼疮 (SLE)三者之间的关系 ,探讨EB病毒在SLE发生发展中的作用机制。方法 用流式细胞仪双色解析法检测SLE患者外周血B淋巴细胞EB病毒潜伏膜蛋白 (LMP1)和抗凋亡基因bc1 2的表达。结果 活动期患者B淋巴细胞LMP1和bc1 2的表达率均高于稳定期患者 ,两组患者均高于对照组 (P均 <0 0 1) ;SLE患者LMP1+ 细胞bc1 2表达率高于LMP1 细胞 (P <0 0 1) ,且SLE患者外周血B淋巴细胞bc1 2的表达与LMP1的表达呈正相关 (r=0 32 8,P <0 0 5 ) ,与病情积分成正相关 (r=0 339,P <0 0 1)。结论 EB病毒感染B淋巴细胞后 ,通过上调受染细胞bc1 2的表达 ,抑制B淋巴细胞的凋亡 ,参与SLE的发生和发展

Abstract:Objective To investigate the relationship among EBV,bc1 2 and systemic lupus erythematosus (SLE),and to elucidate the possible pathogenesis of EBV in the initiation and development of SLE.Methods The expression of EBV LMP1 and bc1 2 in B lymphocytes in peripheral blood of SLE patients was detected by two color cytofluorography.Results The expression of EBV LMP1 and bc1 2 in B cells in active SLE was significantly higher than that in remission, and both were significantly higher than that in control ( P <0 01),The percentage of bcl 2 expression in LMP1 +B cells of SLE patients was significantly higher than that in LMP1 - B cells ( P <0 01),moreover,the percentage of bc1 2 expression was positively correlated with that of LMP1 ( r =0 328, P <0 05) and disease activity ( r =0 339, P <0 05).Conclusion EBV restrains the apoptosis of B lymphocytes by up regulating bc1 2 in the affected B cells and participates in the initiation and development of SLE.
