成人外周血间充质干细胞诱导为神经元样细胞的体外研究

目的:研究成人外周血间充质干细胞分化为神经元样细胞的潜能。方法:体外培养成人外周血中分离出的间充质干细胞,传代纯化后收集第2代细胞。细胞经预诱导及全反式维甲酸再诱导后,通过免疫细胞化学法鉴定诱导后的神经元样细胞。结果:诱导后外周血间充质干细胞分化为具有典型神经元形态的细胞,免疫细胞化学显示细胞神经元烯醇化酶阳性率68%;细胞神经巢蛋白阳性率55%;细胞胶质纤维酸性蛋白表达均阴性。结论:成人外周血间充质干细胞在体外可诱导分化为神经元样细胞。

Differentiation of human Peripheral blood-derived mesenchymal stem cells into neuron like cells in vitro

Objective: To explore differentiation potentiality of human peripheral blood-derived mesenchymal stem cells(PBMSCs)into neuron like cells in vitro.Methods: PBMSCs were isolated from human peripheral blood through density gradient and then amplified.The second passage cells were collected and preinduced,and then induced with ATRA.Neuron like cells were detected by immunocytochemistry.Results: PBMSCs could be differentiated into cells with typical neuronal morphology,68% of the cells expressed NSE,55% of the cells expressed Nsetin,and none expressed GFAP.Conclusion: PBMSCs can be induced to become neuron like cells.
