GRIN1在脑胶质瘤中的表达及其临床意义

摘    要:目的 探讨谷氨酸受体亚基1(GRIN1)在脑胶质瘤中的表达及其临床意义。方法 计算机检索CGGA数据库和GEPIA数据库,下载脑胶质瘤基因表达谱数据和对应的临床数据,采用生信分析方法分析胶质瘤组织GRIN1表达变化及其与胶质瘤病人预后的关系。收集2017年1月至2020年12月手术切除脑胶质瘤标本67例,采用免疫组化染色方法检测GRIN1的表达,病人随访时间截止2021年8月1或病人死亡,记录病人生存情况。结果 生信分析结果显示,胶质瘤组织GRIN1表达水平明显降低(P<0.001),而且,随胶质瘤病理级别增高而明显降低(P<0.001);GRIN1高表达组胶质瘤病人中位生存期较低表达组明显缩短(P<0.0001)。67例胶质瘤分析结果显示,级胶质瘤GRIN1低表达率明显增高(P<0.05),随胶质瘤病理级别增高而明显增高(P<0.05);多因素Cox比例回归风险模型分析结果显示,GRIN1低表达是脑胶质瘤生存预后不良的独立危险因素(P<0.05),生存曲线分析显示,GRIN1低表达组胶质瘤病人中位生存期较高表达组明显缩短(P<0.05)。结论 胶质瘤GRIN1呈低表达,随病理分级增高而明显降低,与胶质瘤病人预后不良的密切相关。

Abstract:Objective To explore the expression of glutamate receptor subunit 1 (GRIN1) in human glioma tissues and its clinical significance. Methods The CGGA and GEPIA databases were searched to download the gene expression data and clinical data of human glioma, and the biometric analysis methods were used to analyze the GRIN1 expression in glioma tissues and its relationship with the prognoses of glioma patients. The expression of GRIN1 was detected by immunohistochemical staining in glioma tissues obtained from 67 glioma patients who underwent microsurgery from January 2017 to December 2020. These 67 glioma patients were followed-up to August 1st, 2021 or the time that the patient died, and the survival status of the patient was recorded. Results The results of biometric analysis showed that the expression level of GRIN1 in glioma tissues was significantly reduced (P<0.001), and it was significantly reduced with the increase of the pathological grade of gliomas (P<0.001); the median survival time of glioma patients in the GRIN1 high-expression group was significantly shorter than the low-expression group (P<0.0001). The analysis results of 67 glioma patients showed that the low expression rate of GRIN1 was significantly increased in glioma tissues (P<0.05), and increased significantly with the increase of the pathological grade of gliomas (P<0.05); low expression of GRIN1 was an independent risk factor for poor survival prognosis of glioma patients (P<0.05); the median survival time of glioma patients in the GRIN1 low-expression group was significantly shorter than the high-expression group (P<0.05). Conclusions The expression of GRIN1 in glioma is low, and it decreases significantly with the increase of pathological grade, which is closely related to the poor prognosis of glioma patients.
