VEGF和COX-2蛋白在脑星形细胞瘤表达的研究

目的 :检测VEGF和COX 2蛋白在脑星形细胞瘤中的表达 ,分析其表达变化与不同病理类型肿瘤的相关性。方法 :选用 5 4例Ⅰ～Ⅳ级星形细胞瘤手术后存档蜡块标本为实验标本 ;应用流式细胞术分别定量检测不同病理类型肿瘤组织中VEGF和COX 2蛋白表达量。结果 :VEGF和COX 2蛋白在星形细胞瘤组织中的表达量 (FI值 )显著增加 ,其表达强度与病理组织学分级显著相关 (P <0 .0 1 )。COX 2蛋白FI值与VEGF蛋白FI值呈显著正相关 (P <0 .0 1 )。结论 :VEGF和COX 2蛋白过度表达在星形细胞瘤形成和演化过程中起重要作用。COX 2蛋白通过诱导VEGF的表达参与肿瘤血管生成。

The expressions of VEGF and COX- 2 protein in astrocytoma

Objective: To investigate the expressions of vascular endothelial growth factor (VEGF) and cyclooxygenase- 2 (COX- 2) protein in astrocytoma,and analyze the correlation between their protein expression quantities and various histopathological grading of tumor. Methods:The experimental group contained 54 paraffin embedded samples from patients with astrocytoma brain tumors. The expressions of VEGF and COX- 2 protein in astrocytoma were detected quantitatively by Flow Cytometry. Results:The FI of both VEGF protein and COX- 2 protein in astrocytoma was higher and correlated with increasing histological grade (all P<0.01).There was positive correlation between the FI of VEGF and the FI of COX- 2 protein (P<0.01). Conclusion:The expressions of VEGF and COX- 2 protein in astrocytoma were high and correlated with the degree of malignancy. These findings indicate that overexpressions of VEGF and COX- 2 protein in tumor cell play an important role in the tumorigenesis and malignant transformation. The expression of COX- 2 protein was correlated with the expression of VEGF protein,indicating that COX- 2 plays a significant role in angiogenesis in astrocytoma through VEGF formation.
