人参总皂苷对老龄大鼠基底前脑-皮质的保护作用

摘    要:目的探讨自由基对基底前脑海马/大脑皮质系统的损害及人参总皂苷(GS)的保护作用,为GS抗脑衰老及改善老龄动物认知能力提供可靠的分子生物学依据。方法Wistar雌性大鼠30只,随机分为3组(n=10):青年组、老龄组、给药组(第17个月始饲以GS至27个月)。采用紫外可见光光度计对3组大鼠基底前脑、海马结构和大脑皮质进行一氧化氮(NO)及丙二醛(MDA)含量测试;同时进行超氧化物歧化酶(SOD)活性测定。结果老龄组基底前脑、海马结构和大脑皮质NO及MDA含量分别较青年组显著增高(P<0.01);但给药组较老龄组降低(P<0.05);老龄组SOD活性显著降低(P<0.01),给药组其活性较老龄组增高(P<0.05)。结论GS具有增强基底前脑海马/大脑皮质系统抗氧化的能力,并能够提高SOD的活性。

Abstract:Objective To investigate the damage of free radicals on the basal forebrain and cortex and the protective functions of ginsenosides(GS),to provide the molecular biological data for the anti-aging effect of GS.Methods Thirty female Wistar rats were randomly divided into 3 groups(n=10):young,aged and GS group(feed up GS to rats from 17th to 27th month).Nitric oxide(NO) and malondialdehyde(MDA) levels as well as superoxide dismutase(SOD) activities were tested in basal forebrain,hippocampal format and cerebral cortex of 3 groups.Results The levels of NO and MDA in the three brain areas were high in aged group(P<0.01) and low in GS group(P<0.05).The activities of SOD were low in aged group(P<0.01) and high in GS group(P<0.05).Conclusions Ginsenosides(GS) can improve the anti-oxidation ability of cholinergic system.
