淫羊藿总黄酮的提取分离工艺研究

目的：考察淫羊藿总黄酮的最佳提取，分离，纯化工艺。方法：采用四因素三水平正交实验进行提取条件的优选，用乙酸乙酯萃取，酸碱沉淀和铅盐沉淀三种分离方法对分离工艺进行比较；分别是用95％乙醇，85％乙醇，正丁醇，乙酸乙酯和甲醇对提取物进行纯化。结果：影响淫羊藿总黄酮提取率的因素依次为溶媒，提取次数，提取时间，提取溶剂的用量，最佳提取工艺为10倍量70％乙醇提取3次，每次1.5h，分离工艺考察表明3种分离方法中以酸碱沉淀法较为理想；提取物经甲醇纯化后总黄酮含量和收率均明显提高，甲醇可作为理想的纯化溶剂。结论：该实验确定的最佳提取工艺稳定性好且简便易行，适合工业化大生产。

STUDIES ON EXTRACTION AND SEPARATION OF TOTAL FLAVONOID OF HERBA EPIMEDII

OBJECTIVE To obtain the best extraction,separation and purification methods of total flavonoid from Herba Epimedii .METHODS The orthogonal design of four factors and three levels were used to elect the optimum extraction method.Three methods as ethyl acetate extraction,acid alkali sedimentation and lead salt sedimentation were compared to determine the most efficient one.RESULTS The factors influenced the extraction efficiency were in following order:solvent,times,time and solvent volume.The optimum extracting condition was to add 10 times amount of 70% ethanol,reflux and extract for 1.5 h,three times totally.It indicated that acid alkail sedimentation was the most efficient and methanol can be used as ideal solvent.CONCLUSION The best extraction process is stable and easy to operate,which is suit to industrialized production.
