摘    要:消炎痛5mg/kg腹腔注射,连续三天,诱导大鼠肝脏金属硫蛋白(MT)含量增加21倍;经消炎痛诱导处理的动物CCl_4中毒时肝脂质过氧化产物丙二醛的形成较对照动物减少32%。消炎痛与离体大鼠肝细胞共同孵育6小时,细胞MT含量增加43%。提示消炎痛诱导肝脏MT含量增加可能是其肝保护作用的重要机制之一。

INDOMETHACIN INDUCED ELEVATION OF HEPATIC METALLOTHIONEIN (MT) SYNTHESIS AND ITS ANTI-INJURIOUS EFFECT ON LIVER WITH CC1_4 INTOXICATION

Abstract:It has been reported that indomethacin may induce protective effect on the liver against toxic injury, the mechanism is not known yet. Our work showed that indomethacin in small dosage of 5mg/ kg/day, i. p. for 3 days induced an increase of rat hepatic MT content as high as 21 times. And in the rats intoxicated with CCl4, the hyperperoxidation product malondialdehyde (MDA) in the liver was 32% less than that in the control group. Furthermore, after the isolated rat hepatocytes were incubated with indomethacin for 6h, their MT content increased by 43%, These results indicated that induction of increased hepatic MT synthesis is probably one of the important mechanisms of protective action of indomethacin on the liver.
