四氢姜黄素抗脂肪肝的实验研究

摘    要:目的观察四氢姜黄素对油酸诱导的正常人肝细胞株L02的体外抗脂肪肝作用及在小鼠体内的抗脂肪肝作用。方法用油酸刺激L02细胞形成脂肪肝细胞模型,以油红O染色镜下观察四氢姜黄素对细胞内脂滴的影响。通过高脂饲料、腹腔注射四环素复合方式建立小鼠脂肪肝动物模型。应用高、中、低剂量组四氢姜黄素进行预防治疗35 d,观察其对肝指数、血脂、肝脂、肝功及肝脏病理的影响。结果四氢姜黄素可使L02细胞内脂滴减少,可降低肝指数、肝内脂质、肝功ALT、AST,能改善肝脏病理形态学,且有明显的量效关系。结论四氢姜黄素具有抗脂肪肝作用,其药效作用强度有一定的量效关系。

Abstract:Objective To observe effects of tetrahydrocurcumin on the hepatocyte steatosis models which was established by treating human L02 liver cells with oleic acid in vitro and fatty liver in mice.Methods The hepatocyte steatosis models were established by treating L02 with oleic acid in vitro to observe the fatty droplets in cells by Oil red O staining under light microscope.The compound fatty liver model of mice was induced by high-fat diet and intraperitoneal injection with tetracycline.The models were preventively treated with tetrahydrocurcumin at high dose,intermediate dose,and low dose respectively for 35 days,and the effects on hepatosomatic index(HIS),blood fat level,liver fat level,liver function,and liver pathology were observed.Results The amount of lipid droplet in L02 cells decreased,HIS,intrahepatic lipid content,and levels of ALT and AST were reduced,and hepatic pathology was improved in significant dose-dependant manner.Conclusion Tetrahydrocurcumin showed effects against fatty liver in dose-dependant manner to some extent.
