单用和复方中5-单硝酸异山梨醇酯的药动学比较研究

摘    要: 目的比较比格犬口服5-单硝酸异山梨醇酯与复方果糖二磷酸钠口服液中的5-单硝酸异山梨醇酯的药动学参数,以研究二者合用对5-单硝酸异山梨醇酯药动学的影响。方法比格犬给药后不同时间采血,用反相高效液相色谱检测血浆中5-ISMN的浓度,用WINBMUPK软件分析数据并计算单用和复方中的5-单硝酸异山梨醇酯的药动学参数,以方差分析和双尾单侧t检验进行比较。结果单用5-单硝酸异山梨醇酯ρmax,AUC0-24h均无明显差异。结论5-单硝酸异山梨醇酯与果糖二磷酸钠组成复方后灌胃给药,对前者的药动学性质无明显影响。

Abstract:OBJECTIVE To compare the pharmacokinetic properties of isosorbide-5-mononitrate (5-ISMN) with 5-ISMN in fluctose-1,6-diphosphate sodium (FDP) compound. METHODS Male Beagle dogs were orally administrated with either 5-ISMN or 5-ISMN in FDP compound. The plasma concentration of 5-ISMN was measured by reversed-HPLC.The data were analyzed and calculated using WINBMUPK software.RESULTS The pharmacokinetic parameters (such as Ka,K,tmax,ρmax,AUC0-24 h) of 5-ISMN and 5-ISMN in FDP compound did not show significant difference.CONCLUSION The use of fluctose-1,6-diphosphate don't influence the pharmacokinetic properties of 5-ISMN.
