摘    要:用McAb亲和层析法粗提HFRS病毒81-14A株感染BHK细胞培养上清液,效果较满意。病毒抗原获得率为48.07%。经McAb亲和层析洗脱,病毒抗原,蛋白峰值基本一致。特异性试验证明洗脱物为HFRS病毒抗原。SDS—PAGE经氨银染色主要有4条区带。

Abstract:HFRS Virus strain 81-14A was propagated in the BHK cells, and the supernatant of infected cell culture was purified roughly by monoclonal antibody affinity chromatography. The result was relatively effective. The recovery of antigens of purified virus was 48.07%. No difference was found in eluting peaks of antigens and proteins of virus. The eluted fractions were identified as antigens of HFRS virus by specific EL1SA tests. SDS-PAGE analysis was carried out by silver stain and four major bands were found. The possibility of rough purification of HFRS virus by immune-adsorbent column with McAb is discussed in this paper.
