| 摘 要： | 目的探讨EphB4在脑胶质瘤细胞中的生物学作用。方法采用免疫组化的方法检测EphB4在脑胶质瘤细胞中的表达。合成EphB4siRNA,应用脂质体转染U251细胞,RT-PCR、Western-blot检测对EphB4的转录及表达的抑制效果,MTT法检测其对细胞增殖的影响,Transwell试验检测细胞侵袭能力,TUNEL法检测细胞凋亡。结果应用siRNA方法明显降低了EphB4的转录(P0.01)及表达(P0.05),明显减慢了细胞的增殖速度(P0.05),加大siRNA浓度后,肿瘤细胞生长抑制进一步增强。S1组(25nmol/L)、S2组(50nmol/L)、S3组(100nmol/L)与U251组、siRNASCR组相比,侵袭能力明显降低(P0.05),凋亡细胞数量有明显增加(P0.05)。结论EphB4在脑胶质瘤细胞的增殖、侵袭、凋亡过程中发挥着重要的生物学调节作用。
