摘    要:目的采用高效液相色谱法同时测定冬虫夏草中尿苷和腺嘌呤2个成分的含量。方法采用Kromasil C18柱(150mm×4.6mm,5μm),以甲醇-水(5:95)为流动相,流速1.0mL/min,检测波长260nm。结果尿苷和腺嘌呤质量浓度质量浓度均在5~500μg/mL范围内与峰面积线性关系良好(r≥0.9991),平均加样回收率分别为99.25%和99.43%,RSD分别为1.18%和1.52%(n=6)。结论该方法快速、准确、重现性好,可用于同时测定冬虫夏草中尿苷和腺嘌呤的含量。

Abstract:Objective To simultaneously determine the contents of uridine and adenine in Cordyceps sinensis by the HPLC method. Methods The analytical column was the Kromasil Cls column(150 mm 4.6 mm, 5 txm). The mobile phase consisted of methanol- water(5:95 ). The flow rate was 1.0 mL/min and the detection wavelength was 260 nm. Results The mass concentrations of uridine and adenine in the range of 5-500 I~g/mL showed the good linear relation with the peak area (r〉0.999 1), and the average recovery rates were 99.25% and 99. 43% respectively. Conclusion This method is quick, accurate, reproducible and can be used for the simultaneous determination of the uridine and adenine contents in Cordyceps sinensis.
