腹部辐射致肠粘膜屏障损伤模型的建立

摘    要:目的根据临床上辐射性肠粘膜屏障损伤防治研究的需要,探讨建立稳定及符合辐射性肠粘膜屏障损伤研究动物模型的方法。方法SD大鼠,分别用8.5Gy或9.5Gy^60Co腹部辐射,光镜、电镜、细菌培养及图象分析进行肠粘膜屏障损伤的评估。结果应用9.5Gy所致的辐射性肠粘膜屏障损害大鼠其肠粘膜形态学、病理学改变,死亡率(33%),及作为肠粘膜屏障损伤的指标之一的肠系膜淋巴结的细菌易位率均稳定,与临床病人的病情接近。结论本模型适合辐射性肠粘膜屏障损伤(如放射性肠炎)治疗(如TPN等)研究的需要。

Abstract:Objective To investigation the right dose for gut mucosa barrier damage model by abdominal radiation(AR) according to the clinical research request. Methods Male SD rats were subjected to an single dosage of 8.5 or 9.5Gy 60 Co AR from the sternen xyphoid to the pubis. The results were analysed according to bacterial culture, light microscope, electric microscope and EMIAS medical colorful image processing and analysis system. Results The mortality of this model at 4th day was approximately 33.4%, and it created a reproducible histological injury which was associated with bacterial translocation to the mesenteric lymph nodes. Conclusions The dose of 9.5Gy was to create a model for research on the therapy of damaged gut mucosa barrier as in radiation enteritis.
