太白贝母与暗紫贝母皂苷类成分的HPLC指纹图谱研究

摘    要:目的:建立不同商品规格太白贝母与暗紫贝母中皂苷类成分的高效液相色谱(HPLC)指纹图谱,为科学评价与有效控制川贝母药材质量提供新方法。方法:色谱柱为Agilent Zorbax SB-C18(250mm×4.6mm,5μm),流动相为乙腈-水(梯度洗脱),检测波长为210nm,柱温为30℃,流速为1.0mL·min-1。结果:建立了太白贝母、暗紫贝母中皂苷类成分的HPLC指纹图谱,标定出21个共有峰;不同商品规格川贝母类药材指纹图谱的相似度在0.95以上,但相对峰面积有不同程度的变化。结论:采用HPLC法建立太白贝母、暗紫贝母皂苷类成分指纹图谱的方法简便、可靠,可为全面有效控制川贝母药材的内在质量提供依据。

Abstract:OBJECTIVE:To establish HPLC fingerprints of saponins from different specifications of Fritillariae taipaiensis and F.unibracteata,and to provide a new method for scientific evaluation and effective quality control of Fritillariae Cirrhosae Bulbus.METHODS:The determination was performed on Agilent Zorbax SB-C 18(250 mm×4.6 mm,5 μm)column with mobile phase consisted of acetonitrile-water(gradient elution)at flow rate of 1.0 mL · min-1.The detection wavelength was set at 210 nm and column temperature was 30 ℃.RESULTS:HPLC fingerprints of saponins in F.taipaiensis and F.unibracteata were established.21 common peaks were identified as the characteristic peaks.The similarity of the fingerprint of saponins from different specifications of Fritillariae Cirrhosae Bulbus was more than 0.95.But the relative peak areas changed to different degrees.CONCLUSION:The method is simple,reliable and provides a basis for effective control of the intrinsic quality of Fritillariae Cirrhosae Bulbus.
