摘    要: 目的考察Ca2+和Ba2+交联果胶凝胶微丸释放蛋白多肽类药物的性质。方法以牛血清白蛋白和阿西美辛为模型药物,分别以Ca2+,Ba2+为交联剂,采用滴制法制备果胶凝胶微丸,考察不同离子交联剂对药物释放的影响。结果在磷酸盐缓冲液6.8中,阿西美辛释放速度比牛血清白蛋白快,果胶钙凝胶微丸释放牛血清白蛋白的速度比果胶钡凝胶微丸快。结论果胶凝胶微丸对于大分子药物牛血清白蛋白具有良好的缓释作用,可通过采用不同离子交联制备获得不同缓释效果,是一种非常有潜力的蛋白药物载体。

Abstract:OBJECTIVE To investigate the swelling properties and release behavior of model drugs(bovine serum albumin and acemetacin,BSA and ACM) from pectin gel beads.METHODS BSA and ACM was as model drugs.Pectinate beads were prepared by the dropping method and crosslinked with Ca2+ or Ba2+.The release and swelling properties were investigated.RESULTS The release rate of ACM from pectin gel beads was faster than BSA and the release rate of BSA from pectin gel beads crosslinked with Ba2+ was slower than that of gel beads crosslinked with Ca2+.CONCLUSION There are significant difference among the pectinate beads crosslinking with different ions.Pectinate gel beads was a potential protein drug vehile which could acquire different release profile by crosslinking with different ions.
