强力霉素对人肺癌细胞系增殖及侵袭行为的影响

摘    要:目的探讨强力霉素对人肺癌细胞系A549细胞的增殖和体外侵袭能力及其基质金属蛋白酶-2(MMP-2)表达的影响。方法采用MTT比色法研究不同浓度强力霉素(2.5、5、10、20、50mg/L)对A549细胞增殖的影响,免疫组织化学方法和Millicell-pore小室法,研究不同浓度强力霉素(2.5、5、10、20mg/L)作用后,A549细胞MMP-2蛋白表达及其体外侵袭转移能力的影响。结果10mg/L以上强力霉素可以抑制A549细胞增殖,5mg/L以上强力霉素可以抑制A549细胞中MMP-2蛋白的表达及体外侵袭。结论强力霉素可以抑制A549细胞增殖及体外侵袭,其抑制A549细胞侵袭及转移的作用与其抑制MMP-2的表达有关。

Abstract:Objective To study the effects of doxycycline on the proliferation,expression of matrix metalloproteinase-2(MMP-2) and the invasiveness of lung cancer carcinoma cell line A549 in vitro.Methods MTT assay was applied to study the effects of doxycycline(2.5,5,10,20,50mg/L) on the prolifera-tion of A549.Immunohistochemistry stain and millicell-pore chamber were used to investigate the expression of MMP-2 and the invasive ability of different concentration of doxycycline(2.5,5,10,20mg/L) treated A549 cells.Results Doxycycline more than 10mg/L inhibited the proliferation of A549 in a nose and time dependent manners.Doxycycline more than 5mg/L inhibited the expression of MMP-2 and invasivenessn of A549.Conclusion Doxycycline can inhibit the proliferation,invasiveness and metastasis of A549 and the mechanism of which is related to the inhibition of MMP-2 in A549 cel1.
