摘    要:探讨神经元型一氧化氮合酶抑制剂(nNOSI)7-硝基吡唑(7-nitroindazole,7-NI)对脑缺血再灌流保护作用的机理。方法选用Wistar大鼠,制成大脑中动脉闭塞模型(MCAO),闭塞1小时后治疗组给予7-NI(50mg/kg),对照和假手术组给予花生油5ml/kg,再灌流1小时后测定各项指标。结果再灌流1小时后,7-NI组NOS(一氧化氮合酶)活性、MDA(丙二醛)含量均明显低于单纯

Abstract:Objective The present study was designed to investigate whether the selective inhibitor of neuronal nitric oxide synthase,7 nitroindazole (7 NI),plays a protective role in the early stage of transient focal cerebral ischemia reperfusion (IR).Methods Wistar rats were divided randomly into 3 groups.The left middle cerebral artery was occluded for 1 hour followed by another hour of reperfusion. 7 NI (50ml/kg) or peanutoil (5ml/kg) was given intraperitoneally just at the beginning of reperfusion.At the end of the experiment,NOS,MDA were measured and the neuronal ultrastructure was evaluated with electron microscope.Results It was found that the activity of NOS and the level of MDA were decreased after ischemia reperfusion in 7 NI group as compared with control group ( P <0.05).The ultrastructural abnormalities were also alleviated in the 7 NI group.Conclusions The results indicate that postischemic application of 7 NI (50mg/kg) protects neurons from focal cerebral IR injury in rats.
