化学修饰的茯苓多糖抗肿瘤效应的组织学观察

摘    要:目的 :揭示化学修饰的茯苓多糖抗肿瘤的效应。方法 :①化学修饰的茯苓多糖 :用 0 .9%NaCl水溶液从茯苓的菌丝体中提取多糖 ,经硫酸酯化 ,获得硫酸酯化的茯苓多糖。②动物实验 :Sarcoma 180肿瘤细胞 (每只 5× 10 6个 )接种于 7~ 8周的BalB/c小鼠的背部皮下 ,然后随机分为茯苓多糖治疗组、5 -Fu阳性对照组和PBS缓冲溶液阴性对照组 ,每组 10只。进行腹腔注射 ,连续注射 7d。③检测 :在最后一次注射后第 2d处死小鼠 ,切取肿瘤 ,称重 ,计算抑瘤率 ,并对肿瘤组织、肝、肾和脾等进行组织学观察。结果 :① 5 -Fu的肿瘤抑制率为 4 6 % ,经过化学修饰的茯苓多糖的抑制率为 38.39%。②组织学观察显示 ,茯苓多糖治疗组可见到肿瘤细胞成片坏死区 ,亦可见核染色质浓缩 ,边聚 ,甚至是半月形 ,说明肿瘤细胞的坏死与凋亡同时存在。③组织学观察还显示 ,经过化学修饰的茯苓多糖对肝、肾无明显的组织学损害 ;与阳性和阴性对照组比较 ,茯苓多糖治疗组小鼠的脾脏脾小节清晰 ,脾中央动脉周围淋巴鞘的淋巴细胞密集 ,表现出对脾的保护作用。结论 :化学修饰的茯苓多糖对小鼠Sarcoma 180肿瘤细胞有杀伤和诱导凋亡的作用 ,并对机体免疫力有增强作用。

Abstract:Objective: To reveal effects of the anti-tumor of the pachyman polysaccharides (PPS) modified by chemical technique. Methods: ① The PPS modified by chemical technique was made from polyglucose which had been abstracted from Poxiacocos by 0.9% sodium chloride. ② 30 BalB/c mice at the age of 7~8 weeks were inoculated with Sarcoma 180 tumorous cells(5×10 6·ml -1) at the hypodermis of the back, and then they were divided into three groups randomly and equally as: the experiment group treated by pachyman polysaccharides,the positive contrast group treated by fluracil and the negative contrast group treated by phosphoric acid buffer. All mice were injected at their bellies once each day and the injection was repeated for six times.③After the mice were killed at the next day to the last time of injection, the tumor was removed and weighed. The inhibition of the tumor was calculated according to the weight of them. In the meantime, the tumor, liver, kidney and spleen were observed histologically. Results: ①The ratio of the inhibition was 46% in the positive contrast group treated by fluracil and it was 38.39% in the experiment group treated by pachyman polysaccharides. ②The histological observation showed the tumorous cells of the experiment group got both necrosis and apoptosis. ③The pachyman polysaccharides modified by chemical technique did no harm to liver and kidney. Moreover, it could protect the spleen. Conclusion: The pachyman polysaccharides modified by chemical technique can kill the Sarcoma 180 tumorous cell of the mouse and induce apoptosis, meanwhile it can improve the immunity of the body.
