南洋参对阿霉素肾病大鼠肾脏保护作用的初步研究

摘    要:目的:观察南洋参对阿霉素肾病大鼠的肾脏保护作用.方法:18只雄性SD大鼠随机分为正常对照组(正常组,n=6)、阿霉素肾病组(模型组,n=6)和南洋参治疗组(治疗组,n=6).模型组和治疗组大鼠一次性尾静脉注射阿霉素7.5 mg/kg诱导阿霉素肾病动物模型,正常组大鼠一次性尾静脉注射等量生理盐水.造模后第1天开始,治疗组大鼠每天以南洋参叶片水煎剂3 ml·500 g-1·d-1灌胃1次,正常组和模型组以蒸馏水3 ml·500 g-1·d-1灌胃,共治疗8周.留取各组大鼠1、4、8周尿标本并行24 h尿总蛋白定量检测,留取第8周时各组大鼠血清及肾脏组织标本,检测肾功能及血清蛋白等生化指标,并观察各组大鼠肾脏病理变化情况.结果:与正常组相比,模型组大鼠24 h尿蛋白定量从第1周开始已明显升高,第4、8周时治疗组较模型组有显著的降低.第8周时,模型组血清白蛋白(Alb)、总蛋白(TP)较正常组有显著降低,治疗组Alb较模型组明显升高,两组间血清总蛋白(TP)差异无统计学意义.模型组血清肌酐(Scr)、尿素氮(BUN)较正常组明显升高,治疗组较模型组明显降低.第8周时肾脏病理结果提示:模型组和治疗组较正常组均有明显肾小球硬化及肾小管间质的损伤,但治疗组的病变明显轻于模型组.结论:南洋参可以降低阿霉素肾病大鼠的蛋白尿水平,减轻肾脏病理损伤,延缓肾衰竭的进展,起到一定的肾脏保护作用.

A Preliminary Study on the Renal Protective Effect of Adriamycin Nephropathy Rats Polyscias Fruticosa

Abstract:Objective:The aim of the present study was to investigate the renoprotective role of polyscias fruticosa in adria- mycin nephropathy, nethotls:18 male Sprague - Dawley rats were randomly divided into three groups: control group, model group and treatment group. Experimental adriamycin nephropathy in rats was induced by a single injection of adriamycin at the dose of 7.5 mg/kg through tail vein in model group and treatment group. Control group rats were injected with equal - volume isotonic saline. After adria- mycin injection, the rats were administrated distilled water( control group and model group)or polyscias fruticosa (treatment group) decoction intragastrically once daily for 8 weeks. 24 h urine samples were collected by metabolic cage on week 1,4, 8. Blood samples and renal tissues were obtained and then the biochemistry indexes were detected and renal pathological change was observed on week 8. Results:After 1 week, the 24 h urine protein excretion was significantly higher in model group than in control group. Compared with model group, the proteinuria in treatment group was attenuated significantly on week 4 and 8. At 8 weeks, hypoAlbuminemia were detected in model group rats and the conditions were alleviated by polyscias fruticosa in treatment group. Compared to control group, serum creatinine and urea nitrogen concentration in model group were significantly higher than control group. Compared to model group, serum creatinine and urea nitrogen concentration in treatment group were significantly decreased. At 8 weeks, renal pathological lesions were induced by adriamycin in both model and treatment groups, but the lesions were alleviated in treatment group compared to model group. Conclusion:Polyscias fruticosa exerts a renoprotective effect through attenuating urinary protein excretion, decreasing blood serum creatinine and urea nitrogen and alleviating renal pathological changes in established adriamycin nephropathy rats.
